To learn more, visit www.valohealth.com. - Work as part of a multifunctional team that include experts in biology, in vitro pharmacology, and ADME - Generate and maintain cell lines using aseptic techniques to support the cell biology and ADME efforts of our drug discovery programs. You will expand, maintain, and plate a diverse number of mammalian cell lines at any given time (including adherent/suspension immortalized cells, primary cells, stable cell lines, etc.). - Perform DNA/RNA transfections and viral transductions in a BSL-2 facility to establish in vitro cell models - Characterize the cell lines you generate, utilizing common techniques such as DNA/RNA isolation, PCR, qRT-PCR, siRNA knockdown, western blot/protein analysis, immunocytochemistry, ELISA, and cell viability/cytotoxicity assays - Optimize the maintenance conditions of a cell line (such as media selection, passages, freeze-thaws, gene knockdown efficiency, protein-of-interest expression levels, etc.) to enable the highest quality data of downstream applications - Keep detailed electronic records of both your work and the cellular stocks in liquid nitrogen - Adhere to safety regulations and good laboratory practices - At least a bachelor’s degree in a scientific discipline with 6+ years of relevant laboratory experience - Demonstrated experience in all steps of cell line generation, from initial genome modifications and clonal selections to cellular characterization and scale up - Proficiency using machines common in these workflows, such as a multidrop Combi, Apricot, Echo, thermocycler, cell counter, fluorescence microscope, and plate reader systems - Experience working with many types of mammalian cell lines (experience with iPSCs and iPSC differentiation is a plus) - Experience with CRISPR-Cas9 workflows, co-culture, organoids, or isolation of primary cells (including hepatocytes) is a plus - Strong written and oral communication skills and a demonstrated ability to convey information to program team members of varying backgrounds - Strong time management skills, with the ability to balance experiments for multiple programs and stakeholders - Ability to troubleshoot, think critically and independently, with excellent attention to detail
